Clayton South is a residential and industrial suburb 20 km south-east of central Melbourne within the local government of Kingston City.
Its surrounding suburbs include Oakleigh South, Clarinda, Heatherton, Clayton, Dingley Village, Mulgrave, Springvale, and Springvale South.
Originally a coastal heathland, the suburb was settled by John O'Shannessy in the early 1840s. He obtained a squatting permit to occupy a 40,000-acre (160 km2) block that included the current suburbs of Clarinda, Clayton South, Dingley, and Heatherton.
Clarinda is located directly west of Clayton South. It was considered to be a part of Clayton South from the 1960s through the 1980s, but in the late 1990s it began to stand alone as a suburb. Thus, Clayton South is situated halfway between Springs Road and Westall Road.
Significant development took place in the suburb after the war, and there was rapid growth in the 1960s and 1970s.
